For the time being, you can get close to 2:1 macro with a 1:1 macro lens and extension tubes. But if you do get tubes, get good ones. canon or kenko for the canon.

if you are considering getting an MP-E 65, then there is no question about it - it should be Canon, btw MP-E is a manual only lens, there is no AF option. As it's been said above, it's a strict macro only lens and it wouldn't be my first choice for macro shooting, it starts at 1:1 and is of no use for anything, but macro.
It takes some time to get used to, and either 60mm or 100mm macro lens will be a better option to start with, you will need an off-camera flash with a TTL cord, diffuser and some sort of flash bracket as well. Adding a set of extension tubes opens up your options as well. Good luck with your choice, tbh you can't go wrong with any true macro lens, Canon, Nikon, Sigma or Tamron will bring you great results and optically they are all very good.
I use Canon 40D with 60mm/2.8, 25mm extension tube, MP-E 65mm/2.8 and Canon 430ex and 270ex flashes for macro and happy with this combo, but MT-24EX twin lite is somewhere on the shopping list
