Hi guys
can anyone comment about the difference b/w
Canon EF-S 60mm f/2.8 MACRO USM LENS and
EF 50mm f2.5 MACRO
regarding optics and image quality excluding the USM part.

I am intending to buy a macro lens for my Digital Rebel XT which I also want to use for potrait purpose. I know Canon EF-S 60mm f/2.8 MACRO USM is new and better, but does it really make a good diffrence to choose it, as its around 100£ more than EF 50mm f2.5 MACRO.

Thanks in advance.

The obvios difference is the 60 is an EFs lens and the 50 is an EF. The 60 is 1:1 and the 50 is not and requites an adapter to capture life size. You should consider third party lenses when you are shopping for a macro. There are some really nice offering out there. Since you want to use yours for a portrait lens I would look also ot the Sigma 50mm. It is a 1:1 and will focus to infinity, a very sharp lens to boot.

... and according to the MTF charts they are rated in the following order:

#1 EF-S 60mm f2.8 Macro

#2 Sigma 50mm F2.8 EX DG

#3 EF 50mm f2.5 compact macro

... note that unlike the other two you can not use the EF-S anywhere but on a cropped digital cameras

I have not tried the 50mm Sigma or Canon. I do have the 60mm and love it. Highly recommended. MTF chart speaks for itself. But then again, EF-S doesn't bother me.
