Is there a way lock up the mirror?

Yup, go to the Camera Setup Menu (the tab with the wrench icon), and then go to the"Mirror lock-up", select "Yes". This is all done using the 4-way direction pad on the back of the camera.

The Mirror Lock Up function is only for cleaning or inspecting the sensor. See page 162 of the manual. You can't take a photo with the mirror locked.
